Randomized studies of retatrutide measure change in body weight as a percentage of baseline, along with absolute weight loss. Glycemic endpoints include hemoglobin A1c and fasting plasma glucose. Investigators also track blood pressure, lipid fractions, and liver fat content to characterize effects beyond weight alone. Trial designs typically use double-blind, placebo-controlled groups with periodic dose escalation, and they record adverse events throughout both treatment and follow-up periods.
Quantification of the peptide in biological samples generally relies on liquid chromatography coupled with tandem mass spectrometry. This approach separates the analyte from matrix components and detects it by mass-to-charge transitions specific to the molecule. Immunoassays offer higher throughput but can cross-react with related peptides and metabolites, so mass spectrometric methods are preferred when structural confirmation is required. Method validation typically addresses accuracy, precision, selectivity, and stability under handling conditions.
Several questions remain unresolved. It is not yet known whether the compound reduces cardiovascular events or mortality, because outcome studies require long follow-up. The durability of weight reduction after treatment withdrawal is uncertain, and rebound has been observed with other incretin-based therapies. Long-term safety data covering several years are limited. Effects in adolescents, in pregnancy, and in people with significant kidney or liver impairment have not been characterized in published reports.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.

Identification and purity assessment typically rely on reversed-phase high-performance liquid chromatography, often paired with mass spectrometry. Mass measurement confirms the expected molecular mass and can reveal truncations or modifications. Peptide mapping and sequencing techniques provide sequence-level confirmation when needed. Because related peptide impurities can behave similarly in a single method, orthogonal techniques are usually combined. Reported purity values depend heavily on the method used and should be interpreted with that in mind.
Dissolution behavior depends on the amino acid sequence, the counterion content, and the buffer chosen. Many peptides disperse readily in water or mild aqueous buffers, while others require a small amount of organic co-solvent or a change in pH. Adsorption to plastic and glass surfaces can reduce the concentration of a solution over time, particularly at low concentrations. Filtration before analysis removes particulates, and aliquoting limits repeated freeze-thaw cycles that stress the material.
Identity and purity are established by instrumental methods rather than by appearance. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and yields a purity value, usually expressed as the share of total peak area. Mass spectrometry checks that the observed mass agrees with the mass calculated from the published sequence, while peptide mapping or amino acid analysis adds structural evidence. Water content, counter-ion identity and residual solvents are sometimes reported as well. A certificate of analysis should name the method behind each figure, because results are method-dependent.
Laboratory handling follows the conventions used for other synthetic peptides. Lyophilized material is weighed and dissolved in an aqueous diluent, typically sterile water or bacteriostatic water, using gentle swirling rather than vigorous shaking, because foaming stresses the chain. Solutions are prepared under clean conditions and, where sterility matters, passed through a suitable filter. Working portions are kept small so that stock material is not repeatedly warmed and cooled, a practice that limits both aggregation and gradual loss of activity.
=== Chromatographic methods === In particular, for the analysis of complex mixtures containing glucose, e.g. in honey, chromatographic methods such as high performance liquid chromatography and gas chromatography are often used in combination with mass spectrometry. Taking into account the isotope ratios, it is also possible to reliably detect honey adulteration by added sugars with these methods. Derivatization using silylation reagents is commonly used. Also, the proportions of di- and trisaccharides can be quantified.
=== IDMS standardization effort === One problem with any creatinine-based equation for GFR is that the methods used to assay creatinine in the blood differ widely in their susceptibility to non-specific chromogens, which cause the creatinine value to be overestimated. In particular, the MDRD equation was derived using serum creatinine measurements that had this problem. The NKDEP program in the United States has attempted to solve this problem by trying to get all laboratories to calibrate their measures of creatinine to a "gold standard", which in this case is isotope dilution mass spectrometry (IDMS). In late 2009 not all labs in the U.S. had changed over to the new system. There are two forms of the MDRD equation that are available, depending on whether or not creatinine was measured by an IDMS-calibrated assay. The CKD-EPI equation is designed to be used with IDMS-calibrated serum creatinine values only.
In January 2025, it was estimated that over 90% of Gaza's population had been displaced. Also the United Nations reported in May 2025 that at least 1.9 million people, roughly 90 percent of Gaza's population, had been displaced by the war. Many families had been uprooted repeatedly, in some cases more than ten times, and new evacuation orders have continued to drive further displacement. Families in Gaza largely have no safe place to go. Displacement sites are packed and conditions are unsafe and undignified. Even where space exists, many lack tents or basic shelter materials, and tented areas themselves continue to come under bombardment.

Helium (from Ancient Greek: ἥλιος, romanized: helios, lit. 'sun') is a chemical element; it has symbol He and atomic number 2. It is a colorless, odorless, non-toxic, inert, monatomic gas and the first in the noble gas group in the periodic table. Its boiling point is the lowest among all the elements, and it does not have a melting point at standard pressures. It is the second-lightest and second-most abundant element in the observable universe, after hydrogen. It is present at about 24% of the total elemental mass, which is more than 12 times the mass of all the heavier elements combined. Its abundance is similar to this in both the Sun and Jupiter, because of the very high nuclear binding energy (per nucleon) of helium-4 with respect to the next three elements after helium. This helium-4 binding energy also accounts for why it is a product of both nuclear fusion and radioactive decay. The most common isotope of helium in the universe is helium-4, the vast majority of which was formed during the Big Bang. Large amounts of new helium are created by nuclear fusion of hydrogen in stars. Helium was first detected as an unknown, yellow spectral line signature in sunlight during a solar eclipse in 1868 by Georges Rayet, Captain C. T. Haig, Norman R. Pogson, and Lieutenant John Herschel, and was subsequently confirmed by French astronomer Jules Janssen. Janssen is often jointly credited with detecting the element, along with Norman Lockyer. Janssen recorded the helium spectral line during the solar eclipse of 1868, while Lockyer observed it from Britain.
==== Commercialization ==== The company announced in November 2021 that it aims to achieve mass-manufacturing in 2024 of its silicon batteries, having pouch-cell samples produced by its manufacturing partner EVE Energy sent to be tested by global car makers. The company announced in 2023 that its batteries will be commercially available in VinFast vehicles in 2025. The CEO said that the company does not plan to become a battery manufacturer or supplier, but instead plans to license its technology to major manufacturers or lease dedicated manufacturing capacity from existing suppliers, saying its silicon batteries can be manufactured using existing factories and manufacturing processes. StoreDot demonstrated its batteries in a prototype Polestar 5 sedan in 2024, however the 2025 model was not set to debut with StoreDot batteries but with batteries developed and manufactured by SK On. StoreDot batteries were still in the testing phase in October 2024 and they had not been incorporated into any upcoming vehicle platforms. StoreDot stated in 2026 it needs two more years of development in order to commercialize its product.

==== 20th century ==== In response to concerns about the risks of occupational polonium exposure, quantities of 210Po were administered to five human volunteers at the University of Rochester from 1944 to 1947, in order to study its biological behaviour. These studies were funded by the Manhattan Project and the AEC. Four men and a woman participated, all suffering from terminal cancers, and ranged in age from their early thirties to early forties; all were chosen because experimenters wanted subjects who had not been exposed to polonium either through work or accident. 210Po was injected into four hospitalised patients, and orally given to a fifth. None of the administered doses (all ranging from 0.17 to 0.30 μCi kg−1) approached fatal quantities. The first documented death directly resulting from polonium poisoning occurred in the Soviet Union, on 10 July 1954. An unidentified 41-year-old man presented for medical treatment on 29 June, with severe vomiting and fever; the previous day, he had been working for five hours in an area in which, unknown to him, a capsule containing 210Po had depressurised and begun to disperse in aerosol form. Over this period, his total intake of airborne 210Po was estimated at 0.11 GBq (almost 25 times the estimated LD50 by inhalation of 4.5 MBq). Despite treatment, his condition continued to worsen and he died 13 days after the exposure event. From 1955 to 1957 the Windscale Piles had been releasing polonium-210.

=== High potassium levels === Spironolactone can cause high blood potassium levels. Rarely, this can be fatal. Of people with heart disease prescribed typical dosages of spironolactone, 10 to 15% develop some degree of hyperkalemia, and 6% develop severe hyperkalemia. At a higher dosage, a rate of hyperkalemia of 24% has been observed. An abrupt and major increase in the rate of hospitalization due to hyperkalemia from 0.2 to 11% and in the rate of death due to hyperkalemia from 0.3 to 2.0 per 1,000 between early 1994 and late 2001 has been attributed to a parallel rise in the number of prescriptions written for spironolactone upon the publication of the Randomized Aldactone Evaluation Study (RALES) in July 1999. However, another population-based study in Scotland failed to replicate these findings. The risk of hyperkalemia with spironolactone is greatest in the elderly, in people with renal impairment (e.g., due to chronic kidney disease or diabetic nephropathy), in people taking certain other medications (including ACE inhibitors, angiotensin II receptor blockers, nonsteroidal anti-inflammatory drugs, the antibiotic trimethoprim, and potassium supplements), and at higher dosages of spironolactone.
=== 1 June === Three people, including a mother and her child, were killed in a Russian air attack on Kyiv in the early hours of the day. The victims were from the Desnianskyi District. Reports said that the casualties were trying to access a locked bomb shelter when the attack occurred. Four people were detained in connection to the incident, namely a security guard, the director of a medical facility, his deputy and the first deputy of the district administration. A subsequent audit of Kyiv's bomb shelters found that nearly half of them were unfit for use or closed. Damage was also reported in the Dniprovskyi District. Ten Iskander missiles were intercepted according to the Ukrainian military. In Russia's Belgorod Oblast, eight people were reported to have been injured by Ukrainian shelling in Shebekino. The regional governor, Vyacheslav Gladkov, claimed that Ukrainian forces used Grad rockets. Two others were injured in a drone attack in Belgorod city. The Russian defence ministry claimed it had repulsed three Ukrainian attacks on Belgorod, killed more than 50 Ukrainian soldiers and destroyed four armored vehicles. It said the attacks involved up to 70 "militants", five tanks, four armoured vehicles, seven pick-up trucks and a Kamaz truck. The Ukrainian Ministry of Infrastructure said the Black Sea Grain Initiative had been halted again because Russia had blocked the registration of ships to all Ukrainian ports.
