The peptide backbone is chemically modified to resist rapid enzymatic breakdown in the body. A fatty acid side chain promotes binding to serum albumin, which slows renal clearance and supports an extended circulation time. These modifications allow less frequent administration than would be possible with an unmodified peptide. The precise contribution of glucagon receptor activation to the overall metabolic effect remains an area of active investigation, because glucagon raises glucose while also increasing energy expenditure.
Development has progressed through early- and mid-stage human studies in adults with obesity and with type 2 diabetes. Published phase 2 data reported reductions in body weight and improvements in glycemic markers over the treatment period. No regulatory agency has approved the compound for any indication, and it remains available only within controlled research settings. Whether benefits observed in trials translate into durable outcomes after treatment stops is not yet established.

Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.
Each receptor contributes a different physiological effect. Activation of the GLP-1 receptor slows gastric emptying and reduces appetite signaling in the brain. GIP receptor activity influences insulin secretion and lipid handling, while glucagon receptor stimulation raises energy use and fat oxidation. Combining these pathways is intended to produce weight loss beyond what single- or dual-receptor agonists achieve. Researchers attribute the observed potency to simultaneous engagement of all three targets, though the exact contribution of each receptor to overall effect remains under investigation.
Clinical development has advanced through phase 2 trials in adults with obesity and type 2 diabetes. Reported phase 2 results described substantial average weight reduction over roughly forty-eight weeks of weekly dosing. A phase 3 program is ongoing to confirm efficacy and assess long-term safety. Because the compound has not received regulatory approval, it is not available as a prescription product. Public discussion of retatrutide often conflates trial findings with marketed status, an important distinction when interpreting coverage of the topic.
The three-receptor design places retatrutide in a distinct category relative to older incretin-based therapies. Single agonists target one receptor, and dual agonists target two. Adding a third target broadens the pharmacological footprint and introduces new trade-offs among efficacy, tolerability, and glucose control. How these trade-offs resolve in large trials is a central focus of current research.
Retatrutide is an investigational peptide studied for obesity and type 2 diabetes. It is a single synthetic molecule designed to activate three metabolic receptors simultaneously. The compound belongs to the incretin mimetic family, a group of peptides that imitate gut hormones involved in appetite and glucose control. Its research code is LY3437943, and it remains under clinical study rather than cleared for routine medical use.
Naja christyi (formerly Boulengerina christyi), commonly known as the Congo water cobra or Christy's water cobra, is a species of venomous snakes belonging to the family Elapidae. The species is native to Sub-Saharan Africa. This species was formerly in the genus Boulengerina, but more recent research by Wallach et al. has shown that Boulengerina is actually a subgenus and Boulengerina christyi is a synonym of Naja christyi. This species has no known subspecies.
Serpins are a superfamily of proteins with similar structures that were first identified for their protease inhibition activity and are found in all kingdoms of life. The acronym serpin was originally coined because the first serpins to be identified act on chymotrypsin-like serine proteases (serine protease inhibitors). They are notable for their unusual mechanism of action, in which they irreversibly inhibit their target protease by undergoing a large conformational change to disrupt the target's active site. This contrasts with the more common competitive mechanism for protease inhibitors that bind to and block access to the protease active site. Protease inhibition by serpins controls an array of biological processes, including coagulation and inflammation, and consequently these proteins are the target of medical research. Their unique conformational change also makes them of interest to the structural biology and protein folding research communities. The conformational-change mechanism confers certain advantages, but it also has drawbacks: serpins are vulnerable to mutations that can result in serpinopathies such as protein misfolding and the formation of inactive long-chain polymers. Serpin polymerisation not only reduces the amount of active inhibitor, but also leads to accumulation of the polymers, causing cell death and organ failure.
== Medical use == Pirbuterol is used in asthma for reversal of acute bronchospasm, and also as a maintenance medication to prevent future attacks. It should be used in patients 12 years of age and older with or without concurrent theophylline and/or inhaled corticosteroid.
Around 90% of the protein structures available in the Protein Data Bank have been determined by X-ray crystallography. This method allows one to measure the three-dimensional (3-D) density distribution of electrons in the protein, in the crystallized state, and thereby infer the 3-D coordinates of all the atoms to be determined to a certain resolution. Roughly 7% of the known protein structures have been obtained by nuclear magnetic resonance (NMR) techniques. For larger protein complexes, cryo-electron microscopy can determine protein structures. The resolution is typically lower than that of X-ray crystallography, or NMR, but the maximum resolution is steadily increasing. This technique is still a particularly valuable for very large protein complexes such as virus coat proteins and amyloid fibers. General secondary structure composition can be determined via circular dichroism. Vibrational spectroscopy can also be used to characterize the conformation of peptides, polypeptides, and proteins. Two-dimensional infrared spectroscopy has become a valuable method to investigate the structures of flexible peptides and proteins that cannot be studied with other methods. A more qualitative picture of protein structure is often obtained by proteolysis, which is also useful to screen for more crystallizable protein samples. Novel implementations of this approach, including fast parallel proteolysis (FASTpp), can probe the structured fraction and its stability without the need for purification.

methylglyoxalase, aldoketomutase, ketone-aldehyde mutase, and (R)-S-lactoylglutathione methylglyoxal-lyase (isomerizing). In some instances, the glutathionyl moiety may be supplied by trypanothione, the analog of glutathione in parasitic protozoa such as the trypanosomes. The human gene for this enzyme is called GLO1.
=== Immunosuppression and myelosuppression === Virtually all chemotherapeutic regimens can cause depression of the immune system, often by paralysing the bone marrow and leading to a decrease of white blood cells, red blood cells, and platelets. Anemia and thrombocytopenia may require blood transfusion. Neutropenia (a decrease of the neutrophil granulocyte count below 0.5 billion/litre) can be improved with synthetic G-CSF (granulocyte-colony-stimulating factor, e.g., filgrastim, lenograstim, efbemalenograstim alfa). In very severe myelosuppression, which occurs in some regimens, almost all the bone marrow stem cells (cells that produce white and red blood cells) are destroyed, meaning allogenic or autologous bone marrow cell transplants are necessary. (In autologous BMTs, cells are removed from the person before the treatment, multiplied and then re-injected afterward; in allogenic BMTs, the source is a donor.) However, some people still develop diseases because of this interference with bone marrow. Although people receiving chemotherapy are encouraged to wash their hands, avoid sick people, and take other infection-reducing steps, about 85% of infections are due to naturally occurring microorganisms in the person's own gastrointestinal tract (including oral cavity) and skin. Dental evaluation and treatment before cytotoxic chemotherapy is recommended for reducing the risk of oral and systemic infections during the neutropenic phase.
=== Helium charged sniffer test === In this case the part is pressurized with helium. The mass spectrometer is fitted with a special device, a sniffer probe usually mounted near the location of expected leak points, that allows the sniffer probe to sample air (and tracer gas when confronted with a leak) at atmospheric pressure and to bring it into the mass spectrometer. This mode of operation is frequently used to locate a leak that has been detected by other methods, in order to allow for parts repair at the detected leak points. Modern machines can digitally remove the helium 20 times below the background level and thus it is now possible detect leaks as small as 5·10−10 Pa·m3·s−1 in sniffing mode.
=== Primary packaging === Primary packaging is directly in contact with the food products, creating the ideal headspace for them, while providing protection from external alteration. Additionally, primary packaging, also known as retail packaging or consumer units, is responsible for the marketing aspects of food packaging. Typically, the packaging materials used in the primary level include cardboard cartons, plastic trays, glass bottles and multi-layered structures (Tetra Pak).

In 2014, the BJP led by Narendra Modi won an absolute majority, in the Indian parliament, and expanded it in 2019. The party joined an NDA coalition after losing its single-party majority in the 2024 general election. Modi remains the longest-serving non-Congress prime minister.
The trees can be harvested for their crimson red resin, called dragon's blood, which was highly prized in the ancient world and is still used today. Around the Mediterranean basin it is used as a dye and as a medicine, Socotrans use it ornamentally as well as dyeing wool, gluing pottery, a breath freshener, and lipstick. The root yields a gum-resin, used in gargle water as a stimulant, astringent and in toothpaste. The root is used in rheumatism, the leaves are a carminative. In 1883, the Scottish botanist Isaac Bayley Balfour identified three grades of resin: the most valuable were tear-like in appearance, then a mixture of small chips and fragments, with a mixture of fragments and debris being the cheapest. The resin of D. cinnabari is thought to have been the original source of dragon's blood until during the medieval and renaissance periods when other plants were used instead. Because of the belief that it is the blood of the dragon it is also used in ritual magic and alchemy. The local inhabitants of the city in the Socotra Island used the dragon's blood resin as a cure-all. Greeks, Romans, and Arabs used it in general wound healing, as a coagulant, cure for diarrhea, for dysentery diseases, for lowering fevers. It was also taken for ulcers in the mouth, throat, intestines and stomach. Dragon's blood from D. cinnabari was used as a source of varnish for 18th-century Italian violin-makers. It was also used as tooth-paste in the 18th century. It is still used as varnish for violins and for photoengraving.
A widely cited study in JAMA which synthesized government and MEDLINE reports, and meta-analyses to evaluate risks from methylmercury, dioxins, and polychlorinated biphenyls to cardiovascular health and links between fish consumption and neurologic outcomes concluded that: "The benefits of modest fish consumption (1-2 servings/wk) outweigh the risks among adults and, excepting a few selected fish species, among women of childbearing age. Avoidance of modest fish consumption due to confusion regarding risks and benefits could result in thousands of excess CHD [congenital heart disease] deaths annually and suboptimal neurodevelopment in children."
