This is a working overview of body composition, written for readers who want more than a one-paragraph summary but less than a textbook.

Trial reports for this compound rely on a small set of repeated measures. Body weight is normally expressed as percent change from baseline at a fixed week, with absolute kilograms given secondarily. Glycemic endpoints include HbA1c, fasting glucose, and, in some protocols, continuous glucose monitoring summaries. Imaging endpoints such as MRI-derived proton density fat fraction quantify liver fat. Standardization matters because a percent change and a categorical responder analysis can tell different stories about the same dataset.
Body composition is assessed with dual-energy X-ray absorptiometry or comparable methods, which separate fat mass from lean mass. Reported losses include both compartments, and the ratio between them is a subject of ongoing analysis rather than a settled result. Waist circumference, blood pressure, and lipid panels are collected as supporting measures. Resting energy expenditure and substrate oxidation are measured in smaller mechanistic studies, where glucagon receptor activity is expected to matter. These substudies are typically short and small, so their findings carry wide uncertainty.
Interpretation depends on study phase and duration. Phase 2 programs are powered for weight and safety signals, not for cardiovascular or renal outcomes, which require event-driven designs. Gastrointestinal events such as nausea, diarrhea, vomiting, and constipation are the most frequently reported adverse effects and tend to cluster around dose escalation. Small increases in heart rate have been described. Because follow-up after treatment discontinuation is limited, questions about weight regain and durability are open rather than answered.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.
Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.

Clinical development has advanced through phase 2 trials in adults with obesity and type 2 diabetes. Reported phase 2 results described substantial average weight reduction over roughly forty-eight weeks of weekly dosing. A phase 3 program is ongoing to confirm efficacy and assess long-term safety. Because the compound has not received regulatory approval, it is not available as a prescription product. Public discussion of retatrutide often conflates trial findings with marketed status, an important distinction when interpreting coverage of the topic.
Retatrutide is a synthetic peptide developed as a single molecule that activates three distinct hormone receptors: GLP-1, GIP, and glucagon. The compound carries the internal designation LY3437943 and was engineered by modifying the backbone of glucose-dependent insulinotropic polypeptide. Its sequence incorporates non-natural amino acids and a fatty acid side chain that extends circulation time. The triple-agonist design aims to combine appetite suppression, improved insulin response, and increased energy expenditure in one agent. Published reports describe it as an investigational product rather than an approved medicine.
Each receptor contributes a different physiological effect. Activation of the GLP-1 receptor slows gastric emptying and reduces appetite signaling in the brain. GIP receptor activity influences insulin secretion and lipid handling, while glucagon receptor stimulation raises energy use and fat oxidation. Combining these pathways is intended to produce weight loss beyond what single- or dual-receptor agonists achieve. Researchers attribute the observed potency to simultaneous engagement of all three targets, though the exact contribution of each receptor to overall effect remains under investigation.
Chymopapain is one of the substracts used in chemonucleolysis (a type of percutaneous discectomy). This method was a new proposal to treat primary lumbar intervertebral disc disease using a nonsurgical method. As a matter of fact, the treatment consists on an injection of proteolytic enzymes to dissolve the herniated nucleus pulposus of the intervertebral discs. Purified chymopapain is the main component of the injection, composed basically of 20 mg in five millilitres. It is provided in vials containing 10.000 units of the lyophilized agent with 0.37 mg of disodium edetate, 3.5 mg of cysteine hydrochloride monohydrate and 1.0 mg of bisulfide. All of them work as stabilisers and activators. Sodium hydroxide is in charge of adjusting the PH of the solution. Then, the injection is rehydrated with 5 milliliters of sterile water. A surgeon injects the solution directly into the herniated disc on the spine to dissolve part of it and ease the pain. This process is under fluoroscopic control. Chymopapain is responsible for catalysis, both in vivo and in vitro, a rapid reduction in the viscosity and, as a consequence, the weight of the nucleus pulposus. In fact, it is a depolymerization of the chondromucoprotein and a decrease in the ability of a disk to imbibe fluid. The dose for a single intervertebral disc is 2 to 4 nanokatals, with a maximum dose per patient of 8 nanokatals. Chymopapain injections are normally given under local, rather than general, anaesthesia. This enzyme has been studied by universities departments around the world.
== Premise == A mysterious man who can turn into a gaseous form appears and calls himself "the Human Vapor". The Human Vapor kills a professor on live television and announces that he intends to kill everyone involved with an organization called "the White Center". Detective Kenji Okamoto is tasked with investigating the murders. Meanwhile, Kenji's ex-girlfriend Kyoko Kono runs her own investigation into the White Room's connection with a meteorite crash 27 years in the past and the cleanup of the contaminated crash site. Kyoko and Kenji find themselves crossing paths during their investigations as they unravel the conspiracy.
== Hepatic first-pass == After a water-soluble drug is swallowed, it is absorbed by the digestive system and enters the hepatic portal system. It is carried through the portal vein into the liver before it reaches the rest of the body. The liver metabolizes many drugs, sometimes to such an extent that only a small amount of active drug emerges from the liver to the rest of the circulatory system. This first pass through the liver thus may greatly reduce the bioavailability of the drug. An example of a drug where first-pass metabolism is a complication and disadvantage is in the antiviral drug remdesivir. Remdesivir cannot be administered orally because the entire dose would be trapped in the liver with little achieving systemic circulation or reaching target organs and cells (for example, cells infected with SARS-CoV-2). For this reason, remdesivir is administered by IV infusion, bypassing the portal vein. However, significant hepatic extraction still occurs because of second pass metabolism, whereby a fraction of venous blood travels through the hepatic portal vein and hepatocytes.

=== Surgery === Either Aetius in the sixth century CE or Persian Ali ibn Abbas al-Magusi in 990 CE conducted the first recorded thyroidectomy as a treatment for goitre. Operations remained risky and generally were not successful until the 19th century, when descriptions emerged from a number of authors including Prussian surgeon Theodor Billroth, Swiss surgeon and physiologist Theodor Kocher, American physician Charles Mayo, American surgeons William Halsted and George Crile. These descriptions provided the basis for modern thyroid surgery. Theodor Kocher went on to win the Nobel Prize in Physiology or Medicine in 1909 "for his work on the physiology, pathology and surgery of the thyroid gland".

=== Autism === Compared to placebo, risperidone treatment reduces certain problematic behaviors in autistic children, including aggression toward others, self-injury, challenging behavior, and rapid mood changes. The evidence for its efficacy appears to be greater than that for alternative pharmacological treatments. The risk of rapid weight gain is an important consideration when prescribing it. Some authors recommend limiting the use of risperidone and aripiprazole to those with the most challenging behavioral disturbances to minimize the risk of drug-induced adverse effects. Evidence for the efficacy of risperidone in autistic adolescents and young adults is less persuasive.

=== Magnetic methods === Magnetic field directed self-assembly (MFDSA) allows the manipulation of dispersion and subsequent assembly of magnetic nanoparticles. This is widely used in the development of advanced materials whereby inorganic nanoparticles (NPs) are dispersed in polymers in order to enhance the properties of the materials. The magnetic field technique allows the assembling of particles in 3D by doing the assembly in a dilute suspension where the solvent does not evaporate. It also does not need to use a template, and the approach also improve the magnetic anisotropy along the chain direction.
== Activators and inhibitors == FFAR2 and FFR3 are activated primarily by short-chain fatty acids (SCFAs) that are 2 to 6 carbons in length (see length of fatty acids). In humans, acetic acid, which has 2 carbon atoms, is a strong activator of FFAR2 but very weak activator of FFAR3; propionic and butyric acids, which have 3 and 4 carbons, respectively, are strong activators of both FFAR2 and FFAR3; pentanoic acid, which has 5 carbon atoms, is a weak activator of FFAR2 but strong activator of FFAR3; and hexanoic acid, which has 6 carbon atoms, is a weak activator of FFAR3 but its effect on FFAR2 has not been reported. More recently, the ketone body fatty acid, acetoacetic acid, while not classified as a SCFA, has been shown to activate FFAR2 with a potency similar to acetic and propionic acids. Many drugs have been developed that bind to and regulate FFAR2's activity. 1) MOMBA, Sorbate, and Compound 1 are orthostatic agonists, i.e., they bind to the same site as SCFAs to activate FFAR2. 2) Compound 58 and AZ1729 are positive allosteric agonists, i.e., they bind to FFAR2 at a site different than the orthostatic binding site and do not by themselves alter FFAR2 activity but enhance the ability of SCFAs and other FFAR2 orthostatic agonists to activate FFAR2. 3) CATPB and BTI-A-404 are reverse agonists, i.e., they bind to the same site as SCFAs but induce a response opposite to that induced by SCFAs.4) 4-CMTB and TUG-1375 are classified as FFAR2 agonists but studies are needed to define their binding sites on FFAR2.
In addition, adipose-derived stem cells from both human and animals reportedly can be efficiently reprogrammed into induced pluripotent stem cells without the need for feeder cells. The use of a patient's own cells reduces the chance of tissue rejection and avoids ethical issues associated with the use of human embryonic stem cells. A growing body of evidence also suggests that different fat depots (i.e. abdominal, omental, pericardial) yield adipose-derived stem cells with different characteristics. These depot-dependent features include proliferation rate, immunophenotype, differentiation potential, gene expression, as well as sensitivity to hypoxic culture conditions. Oxygen levels seem to play an important role on the metabolism and in general the function of adipose-derived stem cells. Adipose tissue is a major peripheral source of aromatase in both males and females, contributing to the production of estradiol. Adipose derived hormones include:

A responder analysis counts participants who cross a threshold, such as five or ten percent weight loss. It complements average percent change by showing how widely results are distributed. The two measures can diverge when a subset of participants loses a large amount.
Mechanistic measures such as energy expenditure and liver fat change come from small substudies and should be read as preliminary. Long-term clinical outcomes are not yet available. Average weight loss figures are better supported than explanations for how the loss is achieved.
Endpoints reported at different weeks, in different baseline BMI ranges, or under different dose-escalation schedules are not directly comparable. Studies in type 2 diabetes often show smaller weight changes than studies in obesity without diabetes. Reporting the population alongside the number keeps comparisons honest.
